      Protein compositions of the Pacific oyster extracts were identified through bottom-up mass spectrometry approach. The proteins were separated on an SDS-PAGE gel, cut into small pieces and tryptic digested (Fig 3A). The peptide mixtures were then subjected to LC-MS/MS analysis generating a total of 81831 spectra for the raw extract and 88130 spectra for the heated extract. Mascot searches were conducted against Pacific oyster proteomes, and the identified proteins were validated by Scaffold (version Scaffold_4.7.3) using criteria at least two peptides at a 99.0% probability. In total Scaffold identified 1,086 proteins from raw extract and 130 proteins from the heated extract, with 103 proteins are shared between the two extracts.
